- CD11b (integrin alphaM subunit) combines with CD18 (integrin beta2 subunit) to form the integrin Mac-1, also known as complement receptor 3 (CR3), which mediates adhesion to C3bi and ICAM-1 (CD54).2, 3 Mac-1 is expressed at varying levels on granulocytes, macrophages, dendritic cells, NK cells, and B-1 cells in the peritoneal and pleural cavities.2-8 Mac-1 expression is rapidly up-regulated on neutrophils after activation and in the same time period that CD62L is shed from the cell surface. In addition to its role in binding C3bi on opsonized targets and mediation of the subsequent ingestion process,4, 9 Mac-1 is important as an adhesion molecule in the transendothelial migration of monocytes and neutrophils.10 Applications: • Flow cytometry1 • In vitro blocking 3, 5 • Immunohistochemistry (frozen sections)11 • Immunoprecipitation1, 2, 5 Recommended Dilution: Flow Cytometry: ≤ 0.3ug/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
