- DECR1 (2, 4DiEnol CoA Reductase, mitochondrial) is a 3336kD member of the DECR subfamily, shortchain dehydrogenase/reductase family of molecules. It is a mitochondrial enzyme that is expressed in multiple cell types, particularly those involved in oxidative degradation of fatty acids. Fatty acids provide energy following their degradation by four enzymes that comprise the betaoxidation cycle. The optimal configuration for fatty acids in this cycle is one of saturation. However, many fatty acids that appear before this cycle are unsaturated, and it is necessary to transform them into a suitable configuration for processing. DECR1 converts a two doublebonded substrate into a one doublebonded substrate, which subsequently undergoes isomerization and entry into the betaoxidation cycle. Human DECR1 is 335 amino acids (aa) in length. It contains a mitochondrial targeting sequence (aa134), and a 301aa mature region that possesses one catalytic site (aa57302). There are two potential acetylation sites at Lys110 and Lys230. DECR1 forms homotetramers. One potential isoform variant shows an alternative start site at Met93 coupled to a deletion of aa292335. Over aa233335, human DECR1 shares 87% aa sequence identity with mouse DECR1. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.25ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
