- AJAP1 (Adherens Junction Associated Protein 1; also Shrew1 and Gm573) is a 4447kD transmembrane protein found on the basolateral surface of epithelium. It is presumably expressed by multiple types of epithelium, and participates in cellcell adhesion. Zonula adherens are structures that participate in the generation of cell contact adherens junctions. Such junctions, or complexes, are key to the maintenance of epithelial cell polarity and the maintenance of celltocell contact. Central to this complex is Ecadherin, a transmembrane molecule that makes crucial contacts with cytoskeletal actin via catenins. AJAP1 is an accompaning transmembrane molecule that interacts with Ecadherin and appears to promote the internalization of Ecadherin upon growth factor stimulation. This facilitates the dissolution of adherens junctions with an increase in cell motility. Notably, AJAP1 is also known to complex with CD147 in nonpolar, invasive cells, suggesting that AJAP1 may play a more complex role in cell migration. Mouse AJAP1 is a 412 amino acid (aa) type III (no signal sequence) transmembrane protein. It contains a 284aa extracellular region (aa1284) plus a 106aa cytoplasmic domain (aa306412). Over aa1283, mouse AJAP1 shares 95% and 75% aa sequence identity with rat and human AJAP1, respectively; over aa158283, mouse AJAP1 sequence identity changes little, showing 95% and 78% equality with rat and human AJAP1, respectively. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
