- The protein is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Applications: Suitable for use in ELISA, Western Blot and Neutralization. Other applications not tested. Recommended Dilution: Sandwich ELISA: 8-10ug/ml detects at least 1000pg/well of recombinant human IL-10 when used with biotinylated anti-human IL-10 (I8432-17) as the detection antibody at a concentration of ~0.5-1ug/ml. Indirect ELISA: 4-8ug/ml detects at least 196pg/ml of recombinant human IL-10. Western Blot: 0.25-0.50ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human IL-10 is 2-4ng/lane, under reducing or non-reducing conditions. Neutralization: 38-57ug/ml yeilds one half of the maximal inhibition [ND50] of the biological activity oh human IL-10 (1mg/ml) Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
