4-plex immunostaining of a frozen tissue section from mouse spleen using four biotinylated antibodies in combination with the 4-color StreptaClick®Color kit. The mouse spleen was harvested and incubated in 4% paraformaldehyde/10% sucrose for 30 min at +20⁰C, followed by incubation in 30% sucrose at +4⁰C overnight. The spleen was mounted in OCT compound, frozen down in liquid nitrogen, cut in 10 μm thick sections, and stored at -20⁰C until staining. The frozen tissue section was fixed in acetone (30 seconds in ice cold 50% acetone/50%water + 10 min in ice cold 100% acetone), air dried, and washed in PBS. The section was then immunostained using the 4-color StreptaClick®Color kit in combination with 4 biotinylated antibodies: biotin rat anti-CD3 (clone 17A2) + StreptaClick®Color 488 (green) biotin rat anti-CD11b (clone M1/70) + StreptaClick®Color 542 (white) biotin rat anti-B220 (clone RA3-6B2) + StreptaClick®Color 594 (red) biotin hamster anti-CD11c (clone N418) + StreptaClick®Color 647 (blue) Images were taken with 10x objective with a standard microscope equipped with appropriate set of filters (SpectraSplit®7) for optimal fluorochrome separation.
