- Vascular endothelial growth factor (VEGF-A/VEGF), originally termed vascular permeability factor (VPF), is a key regulator of angiogenesis and vasculogenesis during development and in adulthood. It belongs to the PDGF family, sharing the characteristic cystine knot motif and functioning as a disulfide-linked homodimer.VEGF is widely expressed in various tissues and cell types-including muscle, hepatocytes, osteoblasts, neutrophils, macrophages, and endothelial cells-and its production is induced by hypoxia and cytokines such as IL-1, IL-6, and TNF-A Alpha.During embryogenesis, VEGF drives endothelial cell proliferation, migration, and survival, thereby regulating vessel density and size. Postnatally, it supports endothelial integrity, wound healing, and female reproductive cycling. Pathologically, VEGF enhances vascular permeability and promotes tumor angiogenesis and metastasis. It also contributes to autoimmune conditions like rheumatoid arthritis and multiple sclerosis, making it a target for anti-angiogenic therapies.
