- Interleukin-22 (IL-22), initially identified as an IL-9-inducible gene (IL-TIF), is a member of the IL-10 cytokine family. The human protein is produced as a 179-amino-acid precursor, cleaved to a 147-aa active form, sharing ~22% and ~79% sequence identity with human IL-10 and mouse IL-22, respectively. Its gene is located on chromosome 12q15. IL-22 signals through a heterodimeric cell surface receptor composed of IL-22R1 (CRF2-9) and IL-10R2 (CRF2-4), activating STAT1 and STAT3 pathways. Its activity is further modulated by a soluble inhibitor, IL-22 binding protein (IL-22BP). Primarily produced by immune cells like activated T cells and Th17 lymphocytes, IL-22 acts predominantly on non-immune epithelial and stromal cells. It plays a critical role in mucosal host defense, tissue repair, and the induction of acute-phase proteins. However, it also contributes to the pathogenesis of chronic inflammatory and autoimmune diseases, highlighting its dual role in regulating tissue homeostasis and inflammation.
