- IKK gamma (IKKG Gamma) is a subunit of a high molecular weight IK KappaB kinase (IKK) complex, which is involved in NF-K KappaB activation. The catalysis of IK KappaB kinase (IKK) complex is generally carried out by three tightly associated IKK subunits. IKKA Alpha and IKKB Beta serve as the catalytic subunits of the kinase and IKKG Gamma serves as the regulatory subunit. Activation of IKK depends upon phosphorylation at Ser177 and Ser181 in the activation loop of IKKB Beta (Ser176 and Ser180 in IKKA Alpha), which causes conformational changes, resulting in kinase activation. Activation of the NF-K KappaB pathway by the T-cell lymphotrophic virus Tax protein or by TNF-A Alpha treatment leads to IKKB Beta-dependent phosphorylation of human IKKG Gamma, primarily at Ser376. In mice, mutation of the orthologous residue (Ser369) to alanine leads to enhanced IKKG Gamma-mediated stimulation of IKKB Beta kinase activity.
