This gene encodes an inhibitor of apoptosis protein (IAP)-binding protein. The encoded mitochondrial protein enters the cytosol when cells undergo apoptosis, and allows activation of caspases by binding to inhibitor of apoptosis proteins. Overexpression of the encoded protein sensitizes tumor cells to apoptosis. A mutation in this gene is associated with young-adult onset of nonsyndromic deafness-61. Alternative splicing results in multiple transcript variants encoding different isoforms.

Flow cytometric analysis of Human Diablo expression on HeLa cells. The cells were stained with purified anti-Human Diablo (Filled hisgram), then a FITC-conjugated second step antibody. To demonstrate specificity of staining, the binding by 10339-R006 was blocked by preincubation of the purified antibody with 20ug recombinant human Diablo (Cat: PKSH031669) for 1 hour (Black solid line hisgram). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.
Immunofluorescence analysis of Human Diablo in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-Human Diablo Monoclonal Antibody (1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI for nuclear staining (blue). Positive staining was localized to cytoplasm.
