- IGF-1 LR3 (Long Arginine 3-IGF-1), a modified analog of IGF-1, features a 13-amino acid extension at the N-terminus and the removal of the third amino acid (glutamic acid). These modifications significantly reduce its affinity for IGF-binding proteins (IGFBPs), resulting in enhanced stability and prolonged activity in culture. In cell culture, IGF-1 LR3 is widely used to promote cell growth, proliferation, and differentiation—particularly in myoblasts, chondrocytes, stem cells (e.g., mesenchymal stem cells), and neural cells. It also exhibits anti-apoptotic effects by activating the Akt pathway and suppressing pro-apoptotic proteins, thereby improving cell survival. This is especially critical under serum-free or low-serum culture conditions. In such environments, IGF-1 LR3 can partially substitute for serum to support the growth of specific cell types, including CHO cells and stem cells.
