- The MPC5 cell line is a conditionally immortalized mouse podocyte model developed to enable studies of podocyte differentiation and function under controlled conditions. This cell line was derived from podocytes isolated from the glomeruli of transgenic mice expressing a temperature-sensitive SV40 large T antigen under the control of an interferon-inducible promoter. Under permissive conditions (33°C with interferon), the MPC5 cells proliferate as undifferentiated, Epithelial-like cells. However, when cultured under non-permissive conditions (37°C without interferon), they undergo growth arrest and differentiation, acquiring features of mature podocytes, such as the formation of arborized processes and expression of podocyte-specific markers like synaptopodin and WT-1. MPC5 cells have proven valuable for in vitro studies of podocyte biology, including cytoskeletal reorganization, differentiation processes, and functional responses to stimuli. During differentiation, these cells exhibit rearrangements of actin and microtubular networks that mimic in vivo podocyte morphology, forming complex foot processes critical for the glomerular filtration barrier. Electrophysiological studies have shown that differentiated MPC5 cells respond to bradykinin via the B2 receptor, indicating functional signaling pathways relevant to podocyte physiology. This cell line has also been instrumental in understanding high glucose-induced podocyte injury, where it has been shown that oxidative stress, cytoskeletal damage, and apoptosis occur, paralleling the pathogenesis of diabetic nephropathy. Recent research highlights the role of molecular regulators, such as miR-204-3p, in modulating high glucose-induced dysfunction in MPC5 cells. This microRNA targets the bradykinin B2 receptor, mitigating apoptotic and oxidative damage pathways activated in hyperglycemic conditions. Such findings underscore the utility of MPC5 cells as a robust model for exploring podocyte-related diseases and testing therapeutic interventions.
