- Caspase-9 belongs to a member of the cysteine-aspartic acid protease (Caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits; large and small. Those dimerize to form the active enzyme. Activation of procaspase-9 by Apaf-1 in the Cytochrome C/dATP-dependent pathway requires proteolytic cleavage to geneRate the mature caspase molecule. Once activated; caspase-9 can initiate a caspase cascade involving the downstream executioner caspase-3; -6 and -7.
