Belonging to the TNF-receptor superfamily, CD40 is expressed on the surface of B-cells and antigen presenting cells. Here it interacts with CD154 (also known as CD40 ligand or CD40L) on activated T-cells to stimulate a range of downstream events which include the initiation and progression of both cellular and humoral adaptive immunity. Dysregulation of CD40 signalling has been observed in autoimmune diseases such as type 1 diabetes, inflammatory bowel disease, psoriasis, multiple sclerosis, rheumatoid arthritis and SLE. CD40 is also widely studied for its role in cancer, with agonistic CD40 monoclonal antibodies showing promising anti-tumour effects.
