4-1BB, also known as CD137, belongs to the TNF-receptor superfamily. It is expressed by T-cells and, upon binding to the 4-1BB ligand (known as 4-1BBL or CD137L) on antigen presenting cells, promotes T-cell proliferation and survival. Since the 4-1BB ligand is expressed by various carcinoma cells, blocking the interaction between 4-1BB and 4-1BBL is investigated as a means of treating cancer. 4-1BB is also studied by researchers investigating autoimmune diseases such as collagen-induced arthritis (CIA), multiple sclerosis and systemic lupus erythematosus (SLE).
